Elza Evren et al. developed a humanized mouse model that supports reconstitution of human blood monocyte and lung macrophage compartments after transplantation of human CD34+ hematopoietic stem and progenitor cells (HSPC). They used a humanized mouse model combined with progenitor cell transplantation, fate localization, intravascular cell labeling, and single-cell RNA sequencing to define the individual development of human macrophages from precursors to mature lung macrophages.
Results showed that classical CD14+ CD16- monocytes were identified as circulating precursors of lung tissue monocytes as well as interstitial and alveolar macrophages. In contrast, nonclassical CD14lo CD16+ monocytes were restricted to the pulmonary vascular system and gave rise to a distinct population of pulmonary intravascular macrophages.
