Description
Monocytes are immune cells that form an important bridge between the innate and adaptive immune response. These cells exist in various phenotypes based on cell surface marker expression and participate in the pathobiology of many systemic diseases. CD14 is known as monocyte differentiation antigen on the surface of myeloid lineage. This protein has a major role in immune recognition and reactivation. CD14+ monocytes are perhaps the most readily available precursors used to generate macrophages and dendritic cells, provide an ideal model for the study of macrophage biology and mechanisms.
These human CD14+ monocytes are purified from peripheral blood by removing other cell types using immunomagnetic negative selection, with the highest viability and plating efficiency. Peripheral blood is collected from donors who have screend for Hepatitis B, Hepatitis C, HIV-1, HIV-2, WNV, and CMV.
Isolated cells are characterized flow cytometry to ensure the enrichment of CD14+ cells.
1 x 10^7 cells per vial.
Cells are negative for bacteria, yeast, fungi, and mycoplasma.
Repeated freezing and thawing of cells is not recommended.

This vial is 1×10^7 cells. Who is it best suited for?

A 1×10^7 vial is ideal for pilot studies, method development, single-condition proof-of-concept experiments, or smaller co-culture setups where you want to avoid unused excess cells. It lets you validate stimulation conditions, differentiation timing, or a flow gating strategy before scaling up to larger cell numbers.

Can I split one vial across multiple experiments?

You can split the contents after thaw into multiple assay arms on the same day, but do not refreeze leftover cells. The page explicitly advises against repeated freeze-thaw cycles because they can reduce viability and shift functional phenotypes. Plan a single-day workflow that uses the full vial across replicates/conditions.

What donor screening is performed, and is this "normal" donor material?

Donors are screened for HBV, HCV, HIV-1/2, WNV, and CMV, and the product disease state is listed as normal. This makes it appropriate as a baseline starting material for mechanistic studies and assay development where you want controlled donor status.

  • The 1×10^7 negative-selected CD14+ monocytes were ideal for our pilot study where we needed smaller scale without sacrificing quality
    Thawing was straightforward, recovery was strong, and the low level of contaminating lymphocytes made downstream phenotyping much cleaner. We ran a short M-CSF differentiation followed by polarization and saw crisp marker separation with minimal background. The smaller vial size also helped us avoid waste and budget overruns.

    These negative-selected monocytes performed very well-post-thaw recovery allowed us to proceed with differentiation quickly, and the resulting macrophages showed stable expression profiles in our controls.     The monocytes tolerated the transition nicely: they adhered, differentiated, and remained viable in the device environment.
