Introduction of Macrophage-targeted Nanoparticle Drug Delivery System

As an important cell type in the immune system, macrophages play a vital role in maintaining the stable state of the immune system, and are involved in cancer and inflammation processes. With the development of nanotechnology, the application of nanosystems targeting macrophages to treat inflammatory diseases, cancer, and cardiovascular diseases has ushered in an upsurge of research.

Targeting Macrophages by NPs to Treat Inflammation

When tissue damage occurs, a large number of M1 macrophages gather at the site of the lesion and cause abnormal inflammation. Based on the negative effect of M1 macrophages on inflammation, approaches targeting M1 macrophages in the inflammatory environment with drug-loaded NPs are highly desirable. Meanwhile, macrophages can migrate to lesion sites quickly and directionally where specific chemokines are secreted, which allows them to be used as vehicles for the targeted drug delivery.

Targeting Macrophages by NPs in Cancer Treatment

Tumor-associated macrophages (TAMs) are an M2-like macrophage population that promote tumor growth via angiogenesis and metastasis. By taking advantage of these characteristics, TAMs can be used as a potential target for cancer therapy by nano drugs. Meanwhile, based on the high plasticity of TAMs, reprogramming these cells to achieve a tumoricidal phenotype has become a popular therapeutic strategy. Targeted drugs based on NPs may provide a new approach for the clinical treatment of cancers.

In addition, macrophage targeting might be used for disease diagnosis, especially for malignant lesions, beyond the delivery of TAM-targeting therapeutic drugs by NPs.
