Description
Monocytes are immune cells that form an important bridge between the innate and adaptive immune response. These cells exist in various phenotypes based on cell surface marker expression and participate in the pathobiology of many systemic diseases. CD14 is known as monocyte differentiation antigen on the surface of myeloid lineage. This protein has a major role in immune recognition and reactivation. CD14+ monocytes are perhaps the most readily available precursors used to generate macrophages and dendritic cells, provide an ideal model for the study of macrophage biology and mechanisms.
These human CD14+ monocytes are purified from peripheral blood, with the highest viability and plating efficiency. Peripheral blood is collected from donors who have screend for Hepatitis B, Hepatitis C, HIV-1, HIV-2, WNV, and CMV.
Isolated cells are characterized flow cytometry to ensure the enrichment of CD14+ cells.
5 x 10^7 cells per vial.
Cells are negative for bacteria, yeast, fungi, and mycoplasma.
Repeated freezing and thawing of cells is not recommended.

Can these be used to generate macrophages for tumor-associated macrophage (TAM)-like modeling in vitro?

Yes. Monocyte-derived macrophages are a practical foundation for TAM-like polarization (e.g., using tumor-conditioned media or defined cytokines). To keep the model interpretable, include a "reference macrophage" condition (standard M-CSF) and then layer TAM-like cues. This helps distinguish donor baseline from tumor-driven reprogramming.

How do you recommend documenting macrophage generation for reproducibility?

Record thaw timing, recovery steps, seeding density, cytokine concentrations, media change schedule, and endpoint day. Those parameters explain most cross-run variability in macrophage phenotype.

Are these cells suitable for high-content imaging endpoints after differentiation?

Yes. For imaging, consistency comes from uniform seeding and edge-effect controls. Pre-coat plates if your protocol requires enhanced adherence, and maintain consistent media volumes across wells to stabilize morphology and signal intensity.
