Background
This gene encodes a cytokine that functions in inflammation and the maturation of B cells. In addition, the encoded protein has been shown to be an endogenous pyrogen capable of inducing fever in people with autoimmune diseases or infections. The protein is primarily produced at sites of acute and chronic inflammation, where it is secreted into the serum and induces a transcriptional inflammatory response through interleukin 6 receptor, alpha. The functioning of this gene is implicated in a wide variety of inflammation-associated disease states, including suspectibility to diabetes mellitus and systemic juvenile rheumatoid arthritis. Elevated levels of the encoded protein have been found in virus infections, including COVID-19 (disease caused by SARS-CoV-2).

What sample types and volume does the IL‑6 qPCR kit require?

This kit is described as a sandwich ELISA with qPCR detection and semi‑quantitative analysis, supporting cell culture supernatant, plasma, and serum. It uses 25 µL sample volume, which is helpful when sample is scarce. For best results, run consistent controls across plates and validate dilution linearity so your semi‑quantitative comparisons remain meaningful.

How does storage compare with standard ELISA kits? We need to store components longer-term.

Storage conditions include 4°C, -20°C, and -80°C (follow protocol guidance per component). The expanded storage options are useful for labs batching experiments. To preserve performance, avoid contamination and repeated freeze-thaw cycles, particularly for sensitive reagents. If you plan multi-week runs, aliquot key components after first reconstitution and keep a consistent lot for the entire study.

Can semi‑quantitative IL‑6 results be integrated into a publication with quantitative ELISA data?

Yes, if you clearly define how you interpret the data. Because this kit is labeled semi‑quantitative, it's best to report relative changes (fold differences) under standardized conditions, and to show validation such as dilution linearity and consistency of controls. Many teams pair semi‑quantitative readouts with a quantitative ELISA for selected samples to demonstrate concordance in trends.

  • Very low sample input made IL‑6 tracking feasible in small studies
The 25 µL sample volume was a major advantage for us because we had limited serum from animal studies. We used this kit primarily to compare groups rather than claim exact concentrations, and the semi‑quantitative patterns were consistent across runs. The qPCR readout integrated smoothly with our existing instruments. With good internal controls, it produced reliable trends over time.
  • Flexible storage options helped us batch experiments without quality loss
Our workflow requires storing reagents at different temperatures depending on experiment cadence. This kit's listed storage range (including -80°C) was helpful for long-term planning. We ran multiple small plates over several weeks and maintained stable results by aliquoting reagents and avoiding freeze-thaw cycles. For semi‑quantitative IL‑6 comparisons, it delivered consistent directionality and clear group separation.
  • Strong semi‑quantitative signal separation in plasma after optimization
We focused on plasma IL‑6 changes during inflammation. After choosing an appropriate dilution strategy and including repeat controls, we saw clear separation between treatment groups. The kit is best when you treat it as a comparative tool, not an absolute pg/mL readout. Once we standardized pipetting and timing, run-to-run consistency improved and the results supported our mechanistic conclusions.
