Background
This gene is one of several chemokine genes clustered on the q-arm of chromosome 17. Chemokines form a superfamily of secreted proteins involved in immunoregulatory and inflammatory processes. The superfamily is divided into four subfamilies based on the arrangement of the N-terminal cysteine residues of the mature peptide. This chemokine, a member of the CC subfamily, functions as a chemoattractant for blood monocytes, memory T helper cells and eosinophils. It causes the release of histamine from basophils and activates eosinophils. This cytokine is one of the major HIV-suppressive factors produced by CD8+ cells. It functions as one of the natural ligands for the chemokine receptor chemokine (C-C motif) receptor 5 (CCR5), and it suppresses in vitro replication of the R5 strains of HIV-1, which use CCR5 as a coreceptor. Alternative splicing results in multiple transcript variants that encode different isoforms.

FAQs Customer Reviews Related Products

What should I know about sample volume and plate setup before ordering?

The kit is specified as 96 tests with a pre-coated plate and 25 µL sample volume per well. It's described as a sandwich ELISA with qPCR detection and semi-quantitative analysis, and sample types include cell culture supernatant, plasma, and serum. These specs are useful for planning when sample volume is limited.

How should I interpret "semi-quantitative" CCL5 results for macrophage activation experiments?

In many macrophage activation studies, what matters most is the direction and magnitude of change across conditions, not a single absolute concentration claim. Semi-quantitative kits can be excellent for that use case when you keep handling consistent and include appropriate controls. If your downstream requirement is absolute quantitation, consider confirming key findings with a fully quantitative colorimetric method on a subset of samples.

What storage approach minimizes kit degradation across a long study?

Storage is listed across 4°C, -20°C, and -80°C depending on components, which helps long studies if you follow the protocol's guidance on which reagents belong where. Split sensitive reagents into aliquots if allowed, avoid repeated freeze-thaw cycles, and track open dates carefully. Keeping consistent thawing practices across run days also improves reproducibility and reduces drift.

  • Great option when sample volume is limited and trends matter most
We often had limited macrophage supernatant volume, and the 25 µL requirement made a big difference. The kit gave consistent relative changes across our treatment groups and aligned well with our transcriptional markers. We treated it as a semi-quantitative method and designed our stats accordingly. With internal controls per plate, we achieved reproducible results across multiple weeks.
  • Pre-coated plates improved consistency between operators in our lab
Different users ran the assay in our lab, and pre-coated plates helped reduce variability. We standardized incubation timing and washing technique, and the results were stable across runs. This kit is particularly good for comparative studies-screening conditions, ranking stimuli, or validating pathway effects-where you want dependable directionality without spending excessive sample volume.
  • Strong fit for macrophage studies with well-controlled sample handling
Once we controlled collection timepoints and avoided freeze-thaw cycles, the assay produced clean separation of groups and good reproducibility. The storage flexibility helped us manage reagents across the project. We also liked that the method naturally encouraged a disciplined workflow with controls and consistent processing. Overall, it supported our CCL5 profiling goals effectively.
