Background
The protein encoded by this gene is a cytokine produced primarily by monocytes and to a lesser extent by lymphocytes. This cytokine has pleiotropic effects in immunoregulation and inflammation. It down-regulates the expression of Th1 cytokines, MHC class II Ags, and costimulatory molecules on macrophages. It also enhances B cell survival, proliferation, and antibody production. This cytokine can block NF-kappa B activity, and is involved in the regulation of the JAK-STAT signaling pathway. Knockout studies in mice suggested the function of this cytokine as an essential immunoregulator in the intestinal tract. Mutations in this gene are associated with an increased susceptibility to HIV-1 infection and rheumatoid arthritis.

FAQs Customer Reviews Related Products

I'm measuring IL-10 from different matrices. What sample types does this kit support, and do I need different preparation steps for serum vs. tissue homogenate?

This is a sandwich, colorimetric, quantitative ELISA designed for common biological matrices including cell culture supernatant, plasma, serum, and tissue homogenate. For best accuracy, we recommend matrix-matching your standards when possible and using proper clarification (centrifugation/filtration) to reduce particulates-especially for tissue homogenates-so background and viscosity effects are minimized.

Is the plate pre-coated and what are the practical runtime and volume needs for a typical experiment day?

Yes, the kit uses a pre-coated plate, which shortens setup and improves consistency. The workflow typically takes about 3 hours from loading to reading, and the sample volume is 100 µL per well. This format is convenient for running a full standard curve plus duplicates, and it supports consistent inter-operator performance because coating variability is removed.

How should I store the kit and what is the shelf-life? I'm planning multiple small runs across a few months.

The kit supports storage at 4°C and -20°C (follow the protocol for component-specific handling), and the listed expiry is 6 months under recommended conditions. For multi-run planning, aliquot any reconstituted standards to avoid repeated freeze-thaw cycles, and keep reagents tightly sealed and protected from contamination to preserve signal stability across your study timeline.

  • Reliable quantitative IL‑10 results with low day-to-day variability
We used this colorimetric sandwich ELISA to quantify IL‑10 in macrophage-conditioned media and serum samples. The pre-coated plate made the workflow smooth, and the whole assay fit within an afternoon. The standard curve looked consistent across two operators, and we saw good separation between stimulated and control conditions without needing excessive troubleshooting. Packaging and component labeling were clear, which helped reduce setup errors.
  • Straightforward protocol and strong signal in culture supernatants
Our lab prioritized ease-of-use because we were screening many conditions. This kit's colorimetric readout was convenient and worked well for cell culture supernatants. The sample volume requirement was manageable, and the pre-coated format reduced variability compared with plates we used to coat ourselves. Turnaround time was fast enough to support weekly experiments and iterative optimization of macrophage polarization assays.
  • Good kit for multi-matrix IL‑10 quantification and planning
We ran plasma, serum, and tissue homogenate in parallel. The kit is positioned for multiple sample types, which matched our project needs. We appreciated the 6‑month expiry because we didn't need to rush all samples into one batch. Storage guidance was simple, and with careful sample clarification the background stayed controlled. Overall, it supported a practical multi-month study schedule.
