Does the medium require supplementation with IL-4, IL-13, or other cytokines?

The M2 Macrophage Generation Medium is formulated as a complete, ready-to-use solution that contains the necessary components to drive alternative activation. In many cases, no additional cytokines are required for a robust M2 shift. However, if you are modeling very specific conditions or wish to further fine-tune the phenotype, you may add extra stimuli under your own protocol after initial polarization.

Can this medium be used with primary human monocytes isolated from different donors?

Yes. It works very well with human PBMC-derived monocytes from various donors, although some variation in response magnitude is expected due to biological differences. The medium provides a consistent framework for inducing M2 characteristics, which you can then confirm by marker expression, cytokine measurement, or functional assays.

What practical tips do you have for achieving optimal M2 polarization using this medium?

We recommend starting with healthy M0 macrophages in good condition, seeding at the advised density, and allowing sufficient adherence before switching to M2 generation medium. Maintain consistent timing across replicates, avoid unnecessary mechanical stress, and ensure that culture conditions (temperature, CO₂, and humidity) are stable. Validating the phenotype using both surface markers and functional readouts will help confirm that your setup is properly optimized.
