Does the medium come as a complete solution, or do I need to add extra cytokines such as IFN-γ or LPS?

The M1 Macrophage Generation Medium is supplied as a ready-to-use formulation that includes the necessary components to drive M1 polarization. In many protocols, no additional cytokines are required to establish a strong M1 profile. However, some users may choose to add specific stimuli for customized or intensified polarization, especially when modeling particular disease-relevant conditions.

Is this medium compatible with monocyte-derived macrophages from different donors or species?

Yes. The formulation has been successfully used to induce M1 phenotypes in human monocyte-derived macrophages as well as in certain rodent models. While the exact expression levels of markers and cytokines may differ between donors or species, the overall shift toward a pro-inflammatory state is consistent. We recommend minor empirical adjustments in culture time or seeding density when working with new sources.

Are there any considerations for using this medium in high-throughput or imaging-based assays?

The medium is compatible with standard multi-well culture formats and imaging workflows. To maximize reproducibility, we recommend optimizing seeding density, ensuring uniform distribution of cells, and providing adequate recovery time before polarization. For imaging, it can be useful to transition to imaging-compatible medium after phenotype induction, depending on your microscope and fluorophore requirements.
