Background
This gene encodes a cytokine that functions in inflammation and the maturation of B cells. In addition, the encoded protein has been shown to be an endogenous pyrogen capable of inducing fever in people with autoimmune diseases or infections. The protein is primarily produced at sites of acute and chronic inflammation, where it is secreted into the serum and induces a transcriptional inflammatory response through interleukin 6 receptor, alpha. The functioning of this gene is implicated in a wide variety of inflammation-associated disease states, including suspectibility to diabetes mellitus and systemic juvenile rheumatoid arthritis. Elevated levels of the encoded protein have been found in virus infections, including COVID-19 (disease caused by SARS-CoV-2).

What are the key differences between this competition IL‑6 kit and a sandwich IL‑6 kit in daily use?

This product is a colorimetric, quantitative competition ELISA for IL‑6. Compared with sandwich assays, competition formats can be helpful in certain conditions where two-site binding is challenging. Operationally, you must be especially disciplined with dilution accuracy and mixing because the signal relationship is inverse with analyte amount. Replicates and consistent incubation timing are essential to reduce variability in competition curves.

What sample types and volumes are required? We have limited material.

The kit lists serum and tissue homogenate as sample types, and the sample volume is 50 µL per well, which is relatively sample-sparing. This is helpful when tissue yield is limited. We recommend running dilution checks for tissue homogenate due to viscosity and matrix effects, and clarifying samples to reduce nonspecific background that could compress your dynamic range.

How long does the assay take, and is the plate pre-coated?

The plate is pre-coated, and the assay time is stated as 1-4.5 hours depending on protocol steps. Pre-coated plates reduce variability and save prep time. For best throughput, prepare your standard dilution series carefully, use consistent wash technique, and keep plate handling uniform across wells-competition assays are sensitive to minor timing differences that can shift the curve.

    We liked that only 50 µL per well was needed, since our tissue homogenate samples were limited.
