In practice, conditioned media often provides the cleanest matrix for soluble factors, while lysates can work if the target is present and the lysis buffer is compatible. We recommend clarifying samples, avoiding harsh detergents, and running matrix-matched dilutions to ensure the standard curve behaves similarly in your sample type.
Repeated freeze-thaw cycles can degrade proteins or alter binding behavior, which increases variability. We recommend aliquoting samples into single-use volumes, storing at appropriate temperatures, and thawing on ice. If multiple thaw cycles are unavoidable, validate by comparing fresh versus twice-thawed aliquots in the same run. Consistent handling across all groups is crucial-differences in storage history can look like biology if not controlled.
Precision improves with disciplined technique: run samples in duplicates or triplicates, keep incubation times identical, and avoid inconsistent washing. Place all comparison groups on the same plate when possible, and include an internal control sample across plates if you must split runs.
