Can this supplement be used with both human and rodent macrophages?

Yes, the Macrophage Growth Supplement is designed to be broadly compatible with both human and rodent primary macrophages as well as commonly used macrophage-like cell lines. It can be added to basal media or combined with our complete macrophage media for enhanced support, depending on your experimental requirements.

Does the supplement influence macrophage polarization toward M1 or M2 phenotypes, or is it neutral with respect to activation state?

The supplement is formulated to be as neutral as possible with respect to polarization. Its primary role is to support cell viability and general health rather than to push the cells toward a specific activation state. When combined with appropriate cytokine cocktails or stimuli, macrophages can still be effectively driven toward M1, M2, or other defined phenotypes

What is the recommended storage and handling to maintain the performance of the supplement over time?

We recommend storing the Macrophage Growth Supplement at −20°C for long-term stability. Aliquoting is advisable to avoid repeated freeze-thaw cycles, which can degrade sensitive components over time. Once thawed and mixed into your culture medium, the supplemented medium can usually be stored at 2-8°C for a limited period, as specified in the product documentation. If you plan to use the supplemented medium over an extended time frame, preparing smaller volumes more frequently can help maintain optimal performance in your cell cultures.

  • Our lab uses both primary macrophages and macrophage-like cell lines, and we wanted a single supplement that could support both systems
    This product has done exactly that. We've seen improved recovery after thawing primary cells and more reliable growth curves in our cell lines. Importantly, the supplement doesn't seem to push the cells toward spontaneous activation, so we retain flexibility in designing polarization experiments.
  • The improved robustness has increased our overall throughput
    We tested this supplement in parallel with our old formulation in a series of experiments measuring phagocytosis and cytokine responses. The cultures supplemented with the Macrophage Growth Supplement consistently produced higher viability and cleaner assay signals. It was particularly evident in longer time-course experiments, where the unsupplemented cultures sometimes crashed.
  • This supplement allowed us to standardize our macrophage culture conditions quickly
    After a short optimization phase, we adopted it as our default for all macrophage-based assays. We've since run multiple screening campaigns using the same supplement and have seen very little batch-to-batch variation in cell behavior.
