This product is a sandwich ELISA kit intended for quantitative measurement of CCL2 with a colorimetric readout. Sandwich formats are widely used for soluble proteins like chemokines because they often provide strong specificity by capturing the target with one antibody and detecting with another. For best performance, follow recommended sample handling, run duplicates, and validate dilution linearity in your matrix.
In macrophage workflows, users most often measure CCL2 in cell culture supernatants, but serum/plasma or tissue homogenates can be relevant in vivo. The key is consistent sample processing: remove particulates, avoid repeated freeze-thaw cycles, and pilot a dilution series. A small optimization step up front usually prevents matrix-driven artifacts later and improves confidence in between-group comparisons.
If samples read above the curve, dilute them in the recommended diluent and re-run so the signal falls within range; record the dilution factor for final reporting. If samples are below the curve, concentrate cautiously (if compatible with your matrix) or increase sample input where the protocol allows. For longitudinal macrophage experiments, it's often better to choose a dilution that keeps most samples mid-curve, then standardize that dilution across the study.
