Description
With linker for activation of T cells (LAT)/linker for activation of B cells (LAB) knockout, this product is isolated from the spleen of pathogen-free laboratory C57BL/6 mice after immortalization with a recombinant retrovirus, which contains the v-raf and myc oncogenes. This product is negative in the tests of mycoplasma, bacteria, fungi and yeast. The expression of CD11b is positive by flow cytometry. It is guaranteed to further culture in the conditions provided by Creative Biolabs. Repeated freezing and thawing of cells is not recommended.

What is the difference between this product (JF50) and the LAB knockout (JF49)?

This product (JF50) is a DKO for both LAT and LAB. The other product (JF49) is a single KO for only LAB. You would use this DKO line if you want to completely eliminate signaling through either of these two key adaptor proteins, as they can sometimes have redundant functions.

What is the specific knockout in this cell line?

This is a double knockout (DKO) cell line. Both the Linker for Activation of T cells (LAT) and the Linker for Activation of B cells (LAB) have been knocked out. These are two closely related adaptor proteins involved in various immune receptor signaling cascades.

  • The cells are growing well and the DKO is solid
    This LAT/LAB DKO line is a fantastic tool. We were seeing conflicting data with our single KO lines, and this DKO line proves the redundant function of LAT and LAB in our pathway.
  • The cells were clean, had high viability, and the fact that they are immortalized saves us so much time and effort
    We confirmed the absence of both proteins. They are on the B6 background, which is perfect. My only note is that spleen-derived cells can be a bit trickier to culture, but following the recommended protocol from CB worked perfectly.
  • A very useful and specific cell line
    The DKO of both adaptors gives us a 'clean-slate' background to study non-LAT/LAB signaling. The cells are CD11b positive and healthy. Thumbs up.
