Description
With DAP12 knockout, this product is isolated from the bone marrow of pathogen-free laboratory C57BL/6 mice after immortalization with a recombinant retrovirus, which contains the v-raf and myc oncogenes. This product is negative in the tests of mycoplasma, bacteria, fungi and yeast. The expression of CD11b is positive by flow cytometry. It is guaranteed to further culture in the conditions provided by Creative Biolabs. Repeated freezing and thawing of cells is not recommended.

What is the function of DAP12, and why would I want a knockout line?

DAP12 (also known as TYROBP) is a key signaling adaptor protein that pairs with various activating receptors on immune cells, including macrophages and NK cells. It's involved in inflammation, phagocytosis, and cell activation. A DAP12 knockout macrophage line is an essential tool for studying the specific pathways and receptors that depend on DAP12 signaling.

What control cell line should I use with this product?

We highly recommend using our standard C57BL/6 Bone Marrow Macrophages (Cat# MTS-0922-JF40) as your wild-type control. It is derived from the same mouse strain (C57BL/6) and immortalized using the exact same method.

What is the protocol for thawing and culturing these cells?

A detailed protocol, including recommended culture medium and thawing instructions, is provided on the product datasheet. The cells are shipped frozen on dry ice and must be stored in liquid nitrogen upon arrival.

    We study TREM2 signaling, which is dependent on DAP12. This cell line was exactly what we needed. We've confirmed the lack of DAP12 signaling and the line is working perfectly in our assays.
