Overview

Description
With signal transducer and activator of transcription-2 (STAT2) knockout, this product is isolated from the bone marrow of pathogen-free laboratory 129 mice after immortalization with a recombinant retrovirus, which contains the v-raf and myc oncogenes. This product is negative in the tests of mycoplasma, bacteria, fungi and yeast. The expression of CD11b is positive by flow cytometry. It is guaranteed to further culture in the conditions provided by Creative Biolabs. Repeated freezing and thawing of cells is not recommended.

STAT2 KO macrophages are particularly valuable for projects focusing on type I interferon pathways, antiviral defenses, and innate immune regulation. They are often used side-by-side with STAT1 KO models to dissect the relative contribution of STAT1 and STAT2 to ISG expression and antiviral activity.

How many passages do you recommend after thawing to maintain physiological relevance and stable phenotype?

We generally recommend limiting these macrophages to 2-3 passages after initial recovery. This helps maintain a phenotype that is close to primary cells and reduces the risk of culture-induced drift. The knockout itself is stable, but extended passaging can influence gene expression and functional responses in any primary-like cell system.

Can I request customized formats?

Yes, we can often accommodate customized formats depending on the project requirements. Some customers request pre-plated cells in multiwell plates for imaging-based assays or compound screening. Others need specific cell numbers per vial or additional QC parameters. If you share details about your experimental platform and throughput, we can propose a tailored solution.
